Description
The process of Sleeping Beauty transposition involves a cut-and-paste mechanism, wherein the transposable element “jumps” from one DNA molecule to another. Integration of the transposon into chromosomes serves as the foundation for sustained, potentially permanent, expression of transgenes in transgenic cells and organisms. The mRNA encoding the hyperactive variant of the Sleeping Beauty transposase is described by Mátés et al. (2009).
The SB-100 mRNA, as outlined in a study by Jin Z, et al. (2011) in Gene Therapy, is responsible for encoding the Sleeping Beauty transposase. This transposase, in conjunction with the Sleeping Beauty transposon, forms a DNA plasmid system utilized in therapeutic human cell genetic engineering. This mRNA possesses a Cap1 structure, which is highly efficient in capping. It is fully substituted with N1-methyl-pseudo Uridine to improve expression and decrease immunogenicity. Additionally, the mRNA features a 110A tail in its sequence, resembling a mature mRNA.
