huADAR2DD is a variant of the human ADAR enzyme engineered to have enhanced deaminase activity. It targets adenosine residues within RNA for editing, converting them to inosine and altering the coding potential of RNA transcripts.
The fusion of dPspCas13b-ADAR2DD(E488Q/T375G) allows for targeted editing of adenosine to inosine in RNA when paired with a guide RNA. This mRNA possesses a Cap1 structure, which is highly efficient in capping. It is fully substituted with N1-methyl-pseudo Uridine to improve expression and decrease immunogenicity. Additionally, the mRNA features a 110A tail in its sequence, resembling a mature mRNA.
